Mechanical crushing of samples with wing knives at very high speed dry or in suspension. Well suited for greasy / brittle, moist / fibrous as well as hard / brittle substances up to Mohs hardness of 5. Some customers have used the HO 4/A for grinding freeze dried leaves for the isolation of chloroplasts.

Cleaning traditional homogenizers is performed by running them in a solution containing detergent. This generally works well enough. Problems arise when high nutrient tissue is flung far up into the tip. This generally leads to bacterial contamination.

Homogenizer accessories — Some glass Teflon and glass-glass Dounce homogenizers

Dounce homogenizers are very easy to clean, but they are prone to shear stress that causes air bubbles, or cavitation, that can damage the material being homogenzied. While many have used Dounce homogenizers for isolating organelles from soft tissue, the HO 4A also appears to be good for this purpose.
