Background
Proliferating cell nuclear antigen (PCNA) is a DNA clamp that acts as a processivity factor for DNA polymerase δ in eukaryotic cells and is essential for replication. PCNA is a homotrimer and achieves its processivity by encircling the DNA, where it acts as a sliding clamp to tether the polymerase to the template strand during synthesis. Beyond its role in DNA replication, PCNA also participates in various aspects of DNA repair, chromatin remodeling, and cell cycle control by serving as a platform for the recruitment of other proteins. Its dynamic interactions and post-translational modifications allow it to coordinate multiple processes crucial for maintaining genomic stability.
