Background

The His-tag (also called 6xHis-tag) is one of the simplest and most widely used purification tags, with six or more consecutive histidine residues. It is the most widely used protein affinity tag. Due to its small size of only six amino acids, it rarely interferes with the structure or function of the target protein. The His-tag binds strongly to divalent metal ions such as nickel or cobalt, which are immobilized on a resin, allowing for efficient purification through immobilized metal affinity chromatography (IMAC). This method enables rapid and specific isolation of recombinant proteins from complex mixtures, making it a valuable tool in molecular biology and biochemistry.
